Millipore’s derivatized CPG media provide reactive functional groups for covalent attachment of ligands. The surface of Millipore’s CPG particles, a porous borosilicate material, is modified by reacting silanol groups on the glass surface with suitable, functionalized silanes. Using a proprietary chemistry, the organic groups are bonded then to the glass surface through stable and consistent covalent silicon-oxygen-silicon bonds. Derivatized CPG matrices are developed with little batch-to-batch variability and with unique characteristics for new application areas. Millipore offers its derivatized CPG products in a variety of chemistries and pore sizes.
Glyceryl CPG (GLY-CPG) Matrices
GLY-CPG matrix is produced by reacting CPG particles with a silane compound to produce anonionic, hydrophilic surface. GLY-CPG media has adjacent diol groups on the two terminal carbon atoms in the aliphatic extension arm that enables oxidation to an active aldehyde by sodium periodate. Covalent attachment of amine containing ligands can be achieved on this solid support.

Long Chain Alkylamine CPG (LCA-CPG) Matrices
The GLY-CPG matrix is used as the starting material for the preparation of Millipore’s LCA-CPG media. This derivatized CPG matrix has a 15 Å aliphatic arm terminating in an active primary amine. The extra long extension facilitates the approach and the attachment of bulky ligands, where steric hindrance and surface repulsion pose problems.

Capacity
Chemical loading for derivatized CPG products is measured by the appropriate titration method. The capacity is dependent on pore size, surface area and chemical derivatization method.

Applications
Compared to many organic particles, Millipore’s CPG medium is a rigid, nonabsorptive matrix unaffected by changes in a solvent system or pressure. Both organic and aqueous solvents can be used without altering pore sizes. This chromatography support is impervious to degradation caused by solutions, such as guanidine, corrosive solvents and concentrated acids. These characteristics contribute to an optimal medium for a number of chromatography applications and related processes, including:
Affinity Chromatography
Affinity chromatography is adsorption chromatography on a solid support carrying biospecific ligands. Antibodies, peptides, hormones, enzymes, and other molecules of biological interest, are immobilized on derivatized CPG products and used for isolating substances showing specific interactions with the ligands. Millipore’s ProSep®-A chromatography media, protein A immobilized onto derivatized CPG matrices, is the protein-A adsorbent of choice for the industrial purification of therapeutic antibodies.
Immobilized Enzymes
Immobilized enzymes are becoming more important in the digestion, modification and transformation of various substances, including biological molecules. Compared to other carriers used in enzyme immobilization, users benefit from the CPG matrix’s rigid structure, broad range of pore sizes and chemistries, stability in organic and aqueous media, and ease-of-use in various modes of operation, such as stirred tank, packed column or expanded bed.
Solid-Phase DNA Synthesis

Cleaning and Regeneration
Derivatized CPG matrices can be cleaned and regenerated using organic solvents and denaturing solutions such as sodium dodecyl sulfate, propanol, methanol, ethanol and urea.
