| Quantitation of antibodies in hybridoma supernatants during antibody production has traditionally been performed using
methods such as ELISA, which are time-consuming and labor-intensive. The Guava RapidQuant™ Human IgG assay is a
simple, no-wash, bead-based, 96-well plate assay, which can be completed in <3 hrs with analysis on the Guava EasyCyte™
microcapillary platform. The RapidQuant™ Human IgG kit yields accurate and precise results in the concentration range of
0.5-20 μg/mL using only 7.5 μL of supernatant and can detect and quantify all human IgG subtypes equivalently. In this study,
the accuracy of concentration prediction by RapidQuant™ Human IgG kits was evaluated using supernatants from myeloma
cell lines producing human antibodies. Myoloma cells were seeded in 12 well plates for 24 hrs after which supernatants were
collected. The antibodies were quantitated by two methods: 1) an in-house ELISA method for IgG quantitation, and 2) the
Guava RapidQuant™ Human IgG assay. The IgG concentration values obtained from the two methods showed a very high
correlation. Further, the ranking of clones by antibody production was virtually identical. The Guava RapidQuant™ Human
IgG Assay thus enables accurate antibody quantitation in a simple and rapid format during the screening and developmental
stages of antibody production. |
