Hybridoma screening and antibody quantification on the Guava Microcapillary Cytometry Platform: A case study

Hybridoma screening is a complex process involving determination of antibody specificity, IgG quantification, antibody isotype, antibody affinity and antibody function in the selection of the best clones. Traditionally, these assays are performed using different experimental techniques and multiple instrument platforms, some of which involve long procedures. The Guava EasyCyte™ bench-top cytometer is a 96-well platform which can be utilized to screen both cells and beads and can be an ideal space-, money-, and labor-saving solution for hybridoma researchers. In this collaborative study, mice were immunized with human CD80 and <30 microliters of total supernatant from each resulting hybridoma was subjected to multiple analyses on the EasyCyte platform. The best clones were identified based on multiple criteria. The antigen specificity was obtained using a cell-based Guava Express™ assay, quantitative levels of IgG in supernatant were determined using the Guava RapidQuant™ Mouse IgG assay, and isotype and relative affinity were also ascertained. Using these analyses, 6 top ranking clones from 455 tested clones were identified based on specificity and productivity data. The Guava EasyCyte platform and assays thus empower researchers by providing multiple layers of information in the ranking and selection of the optimal clones.
