Recombinant adenoviruses are widely used as vectors for introducing heterologous genes into mammalian cells. Their use requires lengthy procedures for selection, purification and propagation of the adenovirus and for titer determination. We report here improved methods for adenovirus purification and titration. In our purification method, viral particles are captured on chemically modified membranes. Our specific wash and elution protocols allow for an efficient recovery of high virus titer.

Accurate viral titer determination is important for maintaining consistent stocks, efficient virus production and overall reproducibility of gene transfer experiments. However, the standard plaque assay for determining infectious titers is tedious, takes two to three weeks and often introduces some variability depending on the cell line and passage number. We have now developed an accelerated method for adenovirus titration and detection. Our assay is ELISA based, takes about 2 hours, and produces consistent results. This method can be used to characterize crude virus preparations from cell lysates as well as purified virus.
